Attività

Il laboratorio di Biotecnologie è un laboratorio multidisciplinare che utilizza e sviluppa approcci diversi, dedicati allo studio delle biotecnologie marine. Il laboratorio copre attività di ricerca legate al campionamento, isolamento e mantenimento di specie marine di interesse biotecnologico (soprattutto microalghe), preparazione di estratti chimici, test delle possibili bioattività degli estratti sia su organismi marini che su cellule umane, analisi a livello genico e proteico sia sulle specie marine risultate attive che su cellule umane.

Sistemi per la sperimentazione

• Facilities per analisi molecolari e chimiche applicate alle biotecnologie marine.
• Camera per colture cellulari umane (cappa biologica tipo 1, incubatore per cellule umane, microscopio).
• Camera per crescita algale.
• Camera con 2 Bioreattori.

Analisi

• Isolamento e mantenimento di microalghe a potenziali applicazioni biotecnologiche.
• Coltivazione su piccola (flask), media (10L carboys) e grande scala (fotobioreattori  da 100 L) di microalghe.
• Genotipizzazione di nuove specie mediante sequenziamento di DNA.
• Saggi di attività biologica di estratti, frazioni chimiche e composti puri su specie marine modello e su linee cellulari umane.
• Saggi di attività antiproliferativa e clonogenicità nei confronti di linee tumorali aggressive.
• Saggi di attività antiossidante e anti-infiammatoria su linee cellulari normali.
• Saggi di attività da danni indotti dalla senescenza cellulare (anti-aging).
• Saggi di attività specifica nei confronti di alcune patologie umane croniche (diabete, Alzheimer, morbo di Crohn).
• Studi molecolari a livello genico e proteico per monitorare l’attività specifica esercitata dalle sostanze bioattive su cellule umane (trasfezione retrovirale, PCR array, immunoblot, ELISA).
• Analisi trascrittomica delle specie microalgali.

Strumentazione

• Strumentazione di base per la biologia molecolare e analisi proteica (PCR, centrifughe, apparati elettroforetici, transilluminatore, bagni termostatici, spettrofotometro,  pH-metro, transblot).
• Cappa biologica tipo 2 per colture algali.
• Cappa chimica.
• Microscopi.
• 2 Fotobioreattori (100 L).
